Research Description: 

The Gordon lab studies metabolism and atherosclerotic cardiovascular disease in two primary lines of investigation:

 Mechanisms regulating dietary fat absorption by the intestine.

  • Genes that control chylomicron secretion.
  • The influence of fat absorption on body weight, metabolism, and atherosclerosis.

 

Inflammatory protease activity in the artery wall and suppression by lipoproteins.

  • Natural mechanisms of inflammatory regulation involving lipoproteins.
  • Development of lipoprotein-targeting protease inhibitors.

Research techniques:

  • Mouse models for atherosclerosis and intestinal lipid absorption studies.
  • Cell culture including genetically altered cell lines and transendothelial transport experiments.
  • Genetic manipulation using CRISPR.
  • Confocal microscopy including immunofluorescence.
  • Electron microscopy.
  • Molecular biology: PCR, quantitative real-time PCR, agarose gel electrophoresis, mouse genotyping.
  • Next-generation sequencening: RNAseq gene-expression analysis.
  • Protein expression techniques: SDS-PAGE, western blotting, immunohistochemistry.
  • Mass spectrometry: Proteomics and structural cross-linking.
